Peningkatan Kadar Glutathione (GSH) melalui Ekstrak Daun Tin: Potensi Terapi Tambahan pada Pengelolaan Stroke
Abstract
Stroke can cause a decrease in glutathione (GSH) levels, which plays an important role in maintaining cell health. The purpose of this study was to evaluate the effect of fig leaf extract on GSH levels after stroke induction in white rats (Rattus norvegicus). The study design was a pre-test and post-test with a control group comparing GSH levels between the negative control group, the positive control group, and the group given fig leaf extract at doses of 250 mg/kgBW and 500 mg/kgBW. Rats were divided into four groups, and GSH levels were measured before stroke induction (D+0), after stroke induction (D+0), and after treatment on day 7 (D+7) and day 14 (D+14). Comparative analysis was performed using ANOVA. The results showed that GSH levels in the negative control group decreased significantly after stroke induction (mean = 0.028, p = 0.00), while the positive control group showed a significant increase in GSH levels (mean = 0.180, p = 0.04). The group given 500 mg/kgBW of fig leaf extract showed a significant increase on D+7 (mean = 0.187, p = 0.03) and D+14 (mean = 0.318, p = 0.009). It was further concluded that fig leaf extract, especially at a dose of 500 mg/kgBW, can increase GSH levels after stroke induction, indicating its potential as an adjunct therapy in stroke management.

ABSTRAK
Stroke dapat menyebabkan penurunan kadar glutathione (GSH), yang berperan penting dalam mempertahankan kesehatan sel. Tujuan penelitian ini adalah mengevaluasi efek ekstrak daun tin pada kadar GSH setelah induksi stroke pada tikus putih (Rattus norvegicus). Rancangan penelitian ini adalah pre test and post test with control group yang membandingkan kadar GSH antara kelompok kontrol negatif, kontrol positif, dan kelompok yang diberi ekstrak daun tin dengan dosis 250 mg/kgBB dan 500 mg/kgBB. Tikus dibagi menjadi empat kelompok, dan kadar GSH diukur sebelum induksi stroke (H+0), setelah induksi stroke (H+0), dan setelah perlakuan pada hari ke-7 (H+7) dan hari ke-14 (H+14). Analisis perbandingan dilakukan dengan ANOVA. Hasil analisis menunjukkan bahwa kadar GSH pada kelompok kontrol negatif menurun signifikan setelah induksi stroke (rerata = 0,028, p = 0,00), sementara kelompok kontrol positif menunjukkan peningkatan kadar GSH yang signifikan (rerata = 0,180, p = 0,04). Kelompok yang diberi ekstrak daun tin 500 mg/kgBB menunjukkan peningkatan signifikan pada H+7 (rerata = 0,187, p = 0,03) dan H+14 (rerata = 0,318, p = 0,009). Selanjutnya disimpulkan bahwa ekstrak daun tin, terutama pada dosis 500 mg/kgBB, dapat meningkatkan kadar GSH setelah induksi stroke, yang mengindikasikan potensi sebagai terapi tambahan dalam pengelolaan stroke.
